Product Overview
The Phanta UniFi DNA Polymerase is a next-generation high-fidelity DNA polymerase carefully selected based on the BioSmart platform, featuring ultra-high fidelity and universal primer annealing. Combined with the latest hot-start technology and superior buffer system, this product has high success rates and specificity at specific universal annealing temperatures, enabling stable amplification of various targets. Its fidelity is 220-fold higher than that of Taq polymerase, providing an efficient and convenient solution for PCR experiments requiring highly precise amplification, such as molecular cloning, sequencing, and site-directed mutagenesis.
The 2 × Phanta UniFi Master Mix is a premix that contains Phanta UniFi DNA Polymerase, dNTPs, Mg2+ , and other necessary components for PCR amplification. It only requires the addition of primers and templates for amplification, thereby reducing pipetting operations and improving detection throughput and result stability. This product is applicable for standard templates, crude products, and GC-rich systems (including primers and templates). The amplification products are blunt-ended, subsequently applicable for ClonExpress Ultra One Step Cloning Kit V3 (#C117) and Ultra-Universal TOPO Cloning Kit (#C603).

Ultra-High Fidelity
The fidelity amplification of different polymerases was determined using the next-generation sequencing method (P. Mielinis. et al. Journal of Molecular Biology (2021)). The result shows that the fidelity of Phanta UniFi DNA Polymerase is ~220 times that of Taq DNA Polymerase (Figure 1).

Universal Program
2 × Phanta UniFi Master Mix (Vazyme #P516)/2 × Phanta UniFi Master Mix (Dye Plus) (Vazyme #P526) were used for parallel comparison with Supplier A, Supplier B, and Supplier C (Figure 2). The results show that P516 and P526 have broad Tm com-patibility and outperform Suppliers A, B, and C. They can efficiently amplify multiple fragments of varying lengths and Tm values on a single PCR machine.

Broad Compatibility
P516/P526 can successfully amplify 17.5 kb gDNA, 18.2 kb plasmid DNA, 17.8 kb λDNA, and 15 kb cDNA (Figure 3). The GC content range is wide, from 24% to 79%, and the local GC compatibility ranged from 22% to 100%. It is compatible with direct amplification from crude samples such as E. coli cultures, Pichia pastoris cultures, mouse tail lysates, rice lysates, and whole blood. Additionally, it supports multiplex PCR with up to 17 targets within 2 kb. The results show that the amplification success rate and specificity of P516/P526 are superior to Suppliers A, B, and C.
