Product Overview
2× Phanta Ultra Master Mix is a next-generation high-fidelity DNA polymerase carefully selected based on the BioSmart platform, offering ultra-high fidelity, rapid amplification at 5 seconds/kb, and broad Tm compatibility. Combining the latest hot-start technology and a superior buffer system, this product exhibits high success rates and specificity at universal annealing temperatures, amplifying fragments up to 40 kb in length. Its fidelity is 200 times higher than Taq polymerase, providing an efficient and convenient solution for PCR experiments requiring high-precision amplification, such as molecular cloning, sequencing, and site-directed mutagenesis. The amplified products are blunt-ended and can be directly used in the ClonExpress Ultra One Step Cloning Kit V3 (Vazyme #C117) and Ultra-Universal TOPO Cloning Kit (#C603).

Ultra-High Fidelity
The fidelity amplification of different polymerases was determined using the next-generation sequencing method (P. Mielinis. et al. Journal of Molecular Biology (2021)). The result shows that the fidelity of Phanta Ultra DNA Polymerase is ~200 times that of Taq DNA Polymerase. See Figure 1 above.

Ultra-fast Amplification
For ~15 kb fragments, Phanta Ultra can save up to 7 hours of PCR time! See Figure 2 above.

Universal Program
2 × Phanta Ultra Master Mix (Vazyme #P518)/2 × Phanta Ultra Master Mix (Dye Plus) (Vazyme #P528) were used for parallel comparison with Supplier A, Supplier B. The results show that P518 and P528 have broad Tm com-patibility and outperform Suppliers A, and B. They can efficiently amplify multiple fragments of varying lengths and Tm values on a single PCR machine. See Figure 3 above.
