Performance
- Ultra-high fidelity
The fidelity of various polymerases was determined using next-generation sequencing (P. Mielinis et al. Journal of Molecular Biology [2021]). The results showed that P505 - Phanta Max Super-Fidelity DNA Polymerase has a fidelity 128-fold higher than that of Taq DNA polymerase (Figure 1).
- Excellent compatibility of high GC content
Combined with an optimized reaction buffer and its exceptional processivity, Phanta Max polymerase efficiently amplifies fragments with high GC content. Using human genomic DNA (gDNA) as a template, target fragments of 654 bp, 900 bp, 800 bp, 1,200 bp, 1,400 bp, and 426 bp were amplified, with a GC content of >68% for each amplicon. As can be seen in Figure 2, all fragments were efficiently amplified under the default conditions.
- Stable crude product amplification ability
Compared to traditional high-fidelity polymerases, Phanta Max polymerase exhibits higher template affinity and inhibitor resistance, resulting in superior performance in crude product amplification. Crude product types tested include microorganisms (e.g., bacteria and fungi), various whole blood, cultured cells, plant tissues, animal tissue lysates, and food lysates. As shown in Figure 3, Phanta Max polymerase efficiently amplifies a variety of crude templates.
- Widely applicable to amplification of various fragment lengths
As shown in Figure 4, using human genomic DNA as a template, under default conditions, target fragments of 0.6 kb, 1.0 kb, 2.6 kb, 3.0 kb, 4.0 kb, 5.1 kb, 6.2 kb, 7.1 kb, 8.5 kb, 10.6 kb, 17.8 kb, 20.3 kb, and 21.4 kb can be efficiently amplified.

PRODUCT OVERVIEW
Phanta Max Super-Fidelity DNA Polymerase is an upgraded version of Phanta Super-Fidelity DNA Polymerase. Compared with the previous generation, Phanta Max has added the unique elongation factor, specificity-enhancing factor and plateau phase anti-inhibitor factor, which greatly improves the long-fragment amplification ability, amplification specificity and amplification yield. Phanta Max can efficiently amplify up to 40 kb simple templates (e.g. λDNA, plasmids), 20 kb complex templates (e.g. genomic DNA) and 10 kb cDNA. The amplification error rate of Phanta Max is 128-fold lower than that of conventional Taq DNA Polymerase. In addition, Phanta Max has a good resistance to PCR inhibitors and can be used for direct PCR amplification of bacteria, fungi, plant tissues, animal tissues, and even whole blood samples. Phanta Max contains two monoclonal antibodies inhibiting the 5'→3' polymerase activity and 3'→5' exonuclease activity at room temperature, which enable it to perform hot start PCR with great specificity. Amplification products are blunt-ended, which are compatible with ClonExpress kits (Vazyme #C117) and TOPO cloning kit (Vazyme #C603).
