Product Overview

The Dual-Luciferase Reporter Gene Detection Kit is used to detect gene regulation. This kit reflects luciferase expression levels by transfecting cells with a reporter plasmid and measuring the fluorescence intensity of the luciferin substrate. To achieve dual-luciferase reporter gene detection, firefly luciferase is detected using luciferin as a substrate, and Renilla luciferase is detected using coelenterate as a substrate, while simultaneously inhibiting firefly luciferase activity.

 

The Dual-Luciferase Reporter Gene Detection Kit can sensitively and efficiently detect luciferase expression regulated by gene elements. Typically, transcriptional regulatory elements are cloned upstream of firefly luciferase, or the 3'-UTR regulatory region is cloned downstream of firefly luciferase. Cells are transfected with the appropriate stimulant, and then lysed to measure luciferase activity. The stimulatory-inducing effect of the regulatory element is assessed by luciferase activity. Renilla luciferase is used as an internal control to correct for transfection efficiency, eliminating differences in cell number and transfection efficiency between different wells. Firefly luciferase catalyzes the emission of luciferin at 560 nm, while Renilla luciferase catalyzes the emission of coelenterate at 465 nm.
