Product Overview
This Mouse Fc Assay Kit is used for high-throughput detection of mouse IgG or mFc fusion protein concentrations in cell supernatants or purified samples. The kit contains an antibody (Anti-Mouse IgG-Eu) that recognizes the Fc region of mouse IgG and labels the fluorescent donor Eu; and a mouse IgG labeled with the fluorescent receptor A2 (Mouse IgG-A2).
In solution, Anti-Mouse IgG-Eu binds to Mouse IgG-A2. The fluorescent donor Eu, when excited by 320 nm light, emits light at 620 nm. Eu then excites the fluorescent receptor A2, which emits light at 665 nm—this is the fluorescence resonance energy transfer (FRET) phenomenon. When mouse IgG or mFc fusion protein is added to the solution, they compete with Anti-Mouse IgG-Eu for binding sites, thus interfering with the FRET phenomenon. (See Figure 1.) The concentration of the tested mouse IgG or mFc fusion protein is inversely proportional to the FRET signal value (the intensity ratio of 665 nm to 620 nm).
