Product Overview
The Human Fc Detection Kit (Broad Spectrum) can be used for high-throughput detection of the concentration of human IgG or hFc fusion proteins in cell supernatants or purified samples by high throughput. The kit contains an antibody (anti-human IgG-Eu) that recognizes the human IgG Fc region and is labeled with a fluorescent donor, Eu; and a human IgG (human IgG-A2) labeled with a fluorescent receptor, A2. In solution, anti-human IgG-Eu binds to human IgG-A2. Eu is excited by 320 nm light, emitting light at 620 nm, which in turn excites the fluorescent receptor A2, emitting light at 665 nm – a fluorescence resonance energy transfer (FRET) phenomenon. When human IgG or hFc fusion proteins are added to the solution, they compete with anti-human IgG-Eu for binding, thus disrupting the FRET phenomenon. The concentration of the human IgG or hFc fusion protein to be detected is inversely proportional to the FRET signal value (the intensity ratio of 665 nm to 620 nm).
