Add&Read Human TNF-α is based on TR-FRET technology, also known as "wash-free ELISA". Compared with traditional ELISA, it only requires two steps, saving more than 80% of time and labor.

 

Product Overview

Tumor necrosis factor-α (TNF-α) is a pro-inflammatory cytokine primarily produced by macrophages and monocytes. It can directly kill tumor cells while exhibiting relatively low toxicity to normal cells. TNF-α assembles intracellularly into a non-covalently linked homotrimer and is expressed on the cell surface. It plays a role in normal inflammatory and immune responses by regulating the production of other cytokines, cell survival, and death, thus coordinating tissue homeostasis.

 

The Add&Read TR-FRET Human TNF-α Quantitative Detection Kit (Customized) uses a sandwich assay to detect TNF-α levels. The kit contains a TNF-α standard and two monoclonal antibodies that specifically bind to TNF-α: one conjugated to europium (Eu) (donor, Anti-TNF-α-Eu), and the other conjugated to A3 (receptor, Anti-TNF-α-A3). When both antibodies bind to TNF-α simultaneously, Anti-TNF-α-Eu and Anti-TNF-α-A3 approach each other, resulting in fluorescence resonance energy transfer (FRET) (Figure 1). A fluorescent donor was excited using 320/340 nm excitation light, emitting light at 620 nm. A fluorescent acceptor was excited using 620 nm light, emitting light at 665 nm. The concentration of TNF-α in the sample was proportional to the FRET signal (the ratio of fluorescence intensity at 665 nm to 620 nm).
